A single DNA mutation in certain proteins can shift their behavior from normal to cancerous, complicating the development of targeted therapies that spare healthy cells. This challenge arises because these mutated proteins closely resemble their healthy counterparts, making it difficult to create treatments that can distinguish between the two without causing unintended harm.
Researchers at NYU Langone Health and its Perlmutter Cancer Center have developed a promising biologic drug that specifically targets a mutated cancer protein called HER2 (human epidermal growth factor receptor 2), while sparing the normal version. HER2 mutations, often found on cell surfaces, can cause uncontrolled cell growth when a single amino acid alteration keeps the protein in a continuously active state. In addition, cancers can occur when cells produce excess copies of the HER2 gene, resulting in high levels of the protein. Lead researcher Dr. Shohei Koide, PhD, explains that their goal was to design an antibody capable of recognizing the single amino acid change in HER2, a feat challenging due to its subtlety. Existing treatments, like trastuzumab and pertuzumab, target HER2 overexpression, but struggle to differentiate between normal and mutant forms of the protein, raising the risk of side effects.
Using a protein-engineering technique, the team created antibodies that selectively bind to mutant HER2, verified through cryo-electron microscopy to refine their precision. They further engineered a bispecific T cell engager, combining it with another antibody to activate T cells, ensuring a targeted immune response against cancer cells while sparing normal cells. In early mouse trials, the treatment successfully inhibited tumor growth without adverse effects, though Dr. Koide notes that further studies are required to assess its safety and efficacy in humans.

The Federal Trade Commission's (FTC) ruling from August, aimed at curbing fake reviews and deceptive testimonials, is now in full effect. This rule prohibits the sale or purchase of reviews or testimonials that falsely claim to be genuine, setting strict limitations on how businesses can use consumer feedback in marketing.
During the proposal and comment period from November 2023 to June 2024, the ruling found significant public support. FTC Chair Lina Khan condemned fake reviews as a drain on consumer trust, noting how they mislead people and harm honest businesses. "By strengthening the FTC’s toolkit to fight deceptive advertising, the final rule will protect Americans from getting cheated, put businesses that unlawfully game the system on notice, and promote fair and honest markets," she stated. This final rule targets reviews and testimonials that falsely represent real user experiences, including those generated by AI or individuals without genuine experience with a business's products or services.
The rule makes it illegal for businesses to create, purchase, or disseminate fake reviews, including those from insiders without disclosure. It prohibits companies from incentivizing specific sentiments in reviews and bans insider reviews that lack transparency about the reviewer’s connection to the business. Additionally, the FTC bars companies from manipulating or concealing negative reviews, including tactics like intimidation, unfounded legal threats, or falsely portraying curated positive reviews as fully representative. This ruling aims to uphold consumer trust by ensuring that reviews accurately reflect genuine customer experiences.

Rescuing Miami's Forgotten Dogs

In South Miami-Dade, a nearby Air Force Reserve base, Eddie Alvarez spends each morning traversing a network of dirt roads in his pickup truck. His mission? To care for the countless stray and abandoned dogs that have made this rural area their home. Now retired, Alvarez has dedicated his time and resources to addressing the needs of these animals, many of whom have been left behind by former owners or displaced by hurricanes. He fills food bowls along the roadside, patiently waiting as familiar dogs emerge from the fields, hungry and wary.
Alvarez operates Eddie’s Dogs, a nonprofit organization committed to supporting the welfare of these homeless animals. His work extends beyond feeding; Alvarez provides medical attention, including deworming, vaccinations, and flea treatments. Each day, he cares for around 25 dogs, constrained not by his willingness but by the limited supply of food he has available. The area, often referred to as "Dead Dog Alley," is fraught with hazards for the animals, especially due to frequent traffic accidents that put the dogs at great risk. Alvarez’s compassion drives him to continue this work, undeterred by the challenges of limited resources and the vast need in the community.
Supporters can contribute to Eddie’s Dogs by donating food or medicine, or by fostering or adopting a dog in need. Alvarez also provides an option for humane surrender for those who can no longer care for their pets, helping to prevent these animals from ending up on the streets. Through his Facebook page and website, he shares updates and information on the dogs he cares for, inviting the public to join his mission to give these animals a second chance at life.
